Quantitattive Analysis of Glucose Transporters of Plasma Membrane in the Isolated rat Adipose Cell in Streptozotocin-Induced Diabetic Rats






Abstract
The effect of insulin dependent diabetes mellitus on glucose transport activity and the concentration of glucose transporter in the plasma membranes in adipose cells isolated from streptozotocin induced diabetic rats have been examined.
The results are as follows.
1) Mean plasma glucose level in streptozotocin induced diabetic rats was 390.7¡¾21.2§·/§£, significantly higher than control (107.0¡¾8.6§·/§£).
2) Mean plasma insulin level in streptozotocin induced diabetic rats was 19.1¡¾9.7uU/§¢, significantly lower than control(43.0¡¾14.9uU/§¢).
3) Glucose transporter of plasma membrane was decreased in streptozotocin induced diabetic rats, compared with control (8.5 pmol/§· protein vs 15.0 pmol/§· protein).
4) The dissociation constant was 2.50¡¿ 10 E-7-M in control, and there was no difference between two groups.
In conclusion, devreased level of plasma insulin and depletion of glucose transporter system in adipose plasma membrane might be contributory to the hyperglycemia in streptozotocin induced diabetic rats.
